The Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences (IOCB Prague) is a leading scientific institution in the Czech Republic, recognized internationally. Its primary mission is basic research in the fields of chemical biology and medicinal chemistry, organic and material oriented chemistry, chemistry of natural compounds, biochemistry and molecular biology, physical chemistry, theoretical chemistry, and analytical chemistry. Today, over 800 employees including 160+ PhD students in 50 research and service groups are involved in a broad spectrum of disciplines, which results in more than 250 publications per year, including papers in the most cited prestigious journals such as Nature or Science. The Institute’s emphasis on the interaction between chemistry and other sciences leads frequently to medicinal applications. An integral part of the mission of the Institute is, therefore, to transfer the scientific results into assets which help people to live better lives. Together with the research expertise, a system of technology transfer, IP management and commercialization have intensively been built-up. Through its subsidiary company IOCB Tech, the Institute helps scientists to transform ideas into patents and matches their research results with interests of commercial partners to bring novel ideas to human use or technology market.

🧫 Methyltransferases regulate many biological processes by adding methyl groups to proteins and other biomolecules. Some have become important drug targets, but others remain poorly explored – including METTL21A, a lysine methyltransferase that trimethylates HSP70 family proteins.

Researchers from IOCB Prague have now reported first-in-class small-molecule inhibitors of METTL21A. The study was carried out by Hugo Kocek, Michal Vaško, Milan Dejmek, Eliška Procházková, Michal Šála, Anna Janovská, Marika Matoušová, Alexandra Dvořáková, Evžen Boura, Radim Nencka, and colleagues.

METTL21A is biologically relevant because it modifies heat shock protein family members and has also been found overexpressed in hepatocellular carcinoma, where higher expression correlates with poor clinical outcome.

The team identified compound 6d as a METTL21A inhibitor and used structure–activity relationship studies to develop improved analogues with fluorinated substituents. This led to metabolically stable 6n, also named HK262, with an IC₅₀ of 1.10 μM.

The researchers also solved a crystal structure of METTL21A in complex with 6m / HK474, revealing how the inhibitor binds and providing a basis for structure-guided optimization.

The work establishes a biochemically and structurally validated starting point for studying METTL21A function and for developing future cellularly active chemical tools targeting this enzyme.

🖤 Celosvětový úspěch Holého látek zachránil miliony životů a přinesl v letech 2001–2025 ústavu prostřednictvím licenčních poplatků miliardy korun.

🧪 Carboxylic acids are abundant, stable, and widely available building blocks in organic chemistry. Turning them into reactive radicals through decarboxylation offers an attractive way to form new carbon–carbon bonds under mild conditions.

Researchers from IOCB Prague, the University of Regensburg, and the University of Strathclyde have now developed a photo(electro)chemical strategy for engaging native carboxylic acids in decarboxylative functionalization. The study includes Luigi Dolcini, Lucie Ludvíková, Jakub Krištof, Michał Domański, Mattia Lepori, Soňa Boháčová, Daniel Bím, Tomáš Slanina, and Joshua Philip Barham.

The team applied the method to reactions of aliphatic carboxylic acids with nitrogen-containing heteroarenes, demonstrating a net oxidative process and assessing the reaction scope, scale-up, and green chemistry metrics.

To understand how the reaction works, the researchers combined transient absorption spectroscopy, radical trapping, EPR experiments, and computational analysis. These studies revealed a key role of the protic solvent, which helps activate the excited dibenzo-fused heterocyclic ketone catalyst.

The results point to a solvent-gated excited-state proton relay mechanism, where proton and electron transfer processes work together to activate native carboxylic acids and generate radical intermediates for further functionalization.
